The field of heart transplantation faces many challenges, but few are as substantial as the large discrepancy between the supply and demand of donor hearts. Due to the shortage of available organs, patients in need of a heart transplant face a high risk of waitlist mortality. The research described in this document is intended to address this issue by investigating the use of extended criteria donors.
A proof of concept study was designed to investigate the potential for using a custom designed asanguineous solution and ex-vivo perfusion circuit to preserve and recondition hearts that otherwise do not meet donation criteria to restore them to normal, viable function -- thus expanding the donor pool. The purpose of this project specifically was to design and operate, according to given specifications, a circuit to perfuse an isolated rat heart.
To date, the designed circuit has been successfully used to perfuse a total of five isolated rat hearts. While the proof of concept study remains in its early phase, preliminary results are promising as the hearts exposed to six hours of ex-vivo perfusion with the custom solution show minimal signs of irreversible cellular injury.
